Alterations in the gut microbiota affect stroke outcomes via modulation of T cells, suggesting a gut-brain axis linking commensal microbes with the CNS.
Alfredo Corell's insight:
Commensal gut bacteria impact the host immune system and can influence disease processes in several organs, including the brain. However, it remains unclear whether the microbiota has an impact on the outcome of acute brain injury. Here we show that antibiotic-induced alterations in the intestinal flora reduce ischemic brain injury in mice, an effect transmissible by fecal transplants. Intestinal dysbiosis alters immune homeostasis in the small intestine, leading to an increase in regulatory T cells and a reduction in interleukin (IL)-17–positive γδ T cells through altered dendritic cell activity. Dysbiosis suppresses trafficking of effector T cells from the gut to the leptomeninges after stroke. Additionally, IL-10 and IL-17 are required for the neuroprotection afforded by intestinal dysbiosis. The findings reveal a previously unrecognized gut-brain axis and an impact of the intestinal flora and meningeal IL-17+ γδ T cells on ischemic injury.

Using Global Burden of Disease (GBD) 2013 methodology, the GBD Cancer Collaboration investigators estimate mortality, incidence, years lived with disability, years of life lost, and disability-adjusted life years for 28 cancers in 188 countries by sex from 1990 to 2013.

Conclusions and Relevance Cancer poses a major threat to public health worldwide, and incidence rates have increased in most countries since 1990. The trend is a particular threat to developing nations with health systems that are ill-equipped to deal with complex and expensive cancer treatments. The annual update on the Global Burden of Cancer will provide all stakeholders with timely estimates to guide policy efforts in cancer prevention, screening, treatment, and palliation.

In this review, we will dissect the microbial actors thought to be involved in the HH as well as their immunomodulatory mechanisms as emphasized by experimental studies, with a particular attention on parasites. The Hygiene Hypothesis (HH) attributes the dramatic increase in autoimmune and allergic diseases observed in recent decades in Western countries to the reduced exposure to diverse immunoregulatory infectious agents. This theory has since largely been supported by strong epidemiological and experimental evidence.

University of Pennsylvania cancer and HIV expert Carl June, MD, has been named one of two recipients of the 2015 Paul Ehrlich and Ludwig Darmstaedter Prize for his outstanding work in cancer immunotherapy. Since 1952, the Paul Ehrlich and Ludwig Darmstaedter Prize has been awarded to scientists who have made great advancements in the fields in which Paul Ehrlich worked, in particular immunology, cancer research, microbiology, and chemotherapy.

He is widely recognized as leader of the team responsible for the first successful and sustained demonstration of the use of CAR T cell therapy, an investigational approach in which a patient’s cells are removed through an apheresis process similar to dialysis and modified in Penn's cell and vaccine production facility. Scientists there reprogram the patients’ T cells through a gene modification technique using a viral vector that trains them to recognize specific types of cancer cells. The modified cells – known as chimeric antigen receptor (CAR) T cells – are then infused back into the patient's body, where they multiply, hunt and attack tumor cells.

For example, a variant of a gene known as a HLA-DQA1, which codes for proteins that sort healthy cells from invading viruses and bacteria, was found in nearly 100% of ancient individuals, but in only 36% of modern ones.

The immune system is a complex structure, built over a person’s life in response to environmental conditions. Antibodies, proteins that tag and attack viruses and bacteria, “remember” past invaders, allowing white blood cells to quickly respond during subsequent infections. Because different groups of people encounter different diseases—the European settlers had high exposure to smallpox, measles, and influenza thanks to close contact with livestock—they develop different antibodies. But what about the genes behind the immune system? Could those also change vulnerability to certain diseases?

Los datos confirman que podemos resetear el sistema inmunológico de pacientes con enfermedades autoinmunes, lo cual es muy esperanzador. Pero advertirles de que hay riesgo de muerte es muy duro, y sólo merece la pena en casos muy concretos en que sepamos que la terapia va a funcionar. Debemos seguir investigando”.

Among children at high risk for allergy in whom peanuts had been introduced in the first year of life and continued until 5 years of age, a 12-month period of peanut avoidance was not associated with an increase in the prevalence of peanut allergy. Longer-term effects are not known.

A study showing that vasoactive intestinal peptide (VIP) is able to reduce the pathogenic profile of Th17 cells and favoring Treg cell differentiation.

These results indicate that VIP may have promising effects in the early phase of RA targeting the inhibition of pathogenic Th17 cells,and favoring Treg cell differentiation.

A 5-year-old boy confined to a hospital room due to SCID (a rare immune disorder) is asking people to wear yellow in a show of support before his next operation — and the response on social media has been overwhelming, his family says.
