Fusion protein activates immune cells against tumors in less costly, more accessible way

A novel approach to cancer immunotherapy—strategies designed to induce the immune system to attack cancer cells—may provide a new and cost-effective weapon against some of the most deadly tumors, including ovarian cancer and mesothelioma.

Investigators from Harvard Medical School and the Massachusetts General Hospital Vaccine and Immunotherapy Center report in the Journal of Hematology & Oncology that a protein they engineered was able to prolong survival in animal models of both tumors.  Their success came from combining a molecule that targets a tumor-cell-surface antigen with another protein that stimulates several immune functions.

El primer trasplante renal cruzado en España tuvo lugar en 2009. El procedimiento, que consiste en intercambiar donantes entre parejas incompatibles entre sí, era un intento de profundizar en el camino de obtener órganos de donantes vivos, compensar de esta forma la caída de órganos procedentes de cadáver por la reducción de muertes en accidentes de tráfico y reforzar la buena salud del programa de trasplantes español. Desde entonces, ya se han practicado 51 intervenciones de este tipo, 26 de ellas este mismo año, como ha anunciado la Organización Nacional de Trasplantes (ONT), que considera que cuatro años después, ya se puede decir que el programa se ha consolidado.

First described in 1975, natural killer cells are part of the innate immune system and have been recognized for their ability to kill malignant cells without prior sensitization. Recent studies suggest that these cells may play an important role in human cancer prevention and could be an effective tool in cancer immunotherapy. 

Natural killer cells have been shown to kill transformed or virally infected cells without antigen priming. In addition, natural killer cells are able to protect against graft-versus-host disease.

En mi opinión, a los 12 años una gran parte de la población esta inmunizada de forma natural frente a la varicela por lo que resultaría poco útil vacunar a los niños a los 12 años, aunque se evitarían los casos más graves ya que la gravedad de esta enfermedad se incrementa con la edad y es de gran importancia la inmunidad en las embarazadas. Además creo que se debería replantear el suministrar la vacuna frente al virus del papiloma humano también en los varones pues se ha demostrado su asociación con el cáncer de recto. En definitiva, la realización de un calendario vacunal común para todas las comunidades me parece muy necesario sin embargo creo que se podría mejorar este plan propuesto por Sanidad.

Clinical Context

Although biologic agents have greatly improved outcomes for patients with autoimmune diseases, substantial concern remains about risk for serious infections associated with these drugs. A previous study by Grijalva and colleagues (JAMA. 2011;306:2331-2339) examined 1172 cases of serious infection among patients with rheumatoid arthritis (RA), psoriasis, and inflammatory bowel disease. Pneumonia and skin or soft tissue infection accounted for most cases of infection. Compared with nonbiologic treatment regimens, anti–tumor necrosis factor (TNF) drugs were not associated with a significant increase in the risk for infection in any disease state studied.

Nonetheless, the rate of serious infection among patients with RA treated with biologic agents is estimated to be 3 to 7 infections per 100 person-years. The current study by Curtis and colleagues examined factors that might predict a higher rate of serious infection associated with the use of anti-TNF drugs for RA.

Wahn U et al. – This study confirmed that this sublingual immunotherapy (SLIT) preparation significantly reduced symptoms and medication use in children with grass pollen–allergic rhinoconjunctivitis. The preparation showed significant effects on allergen–specific antibodies, was well tolerated, and appeared to be a valid therapeutic option in children allergic to grass pollen.

De hecho, las unidades de Murcia, Navarra, Zaragoza, Asturias, Valladolid y la del Clínic de Barcelona no han alcanzado esta actividad mínima en los últimos cinco años y algunas, como el Virgen de la Arrixaca murciano, apenas realizó tres trasplantes de corazón en 2011.

In the 1980s and 1990s, the UK outbreak of bovine spongiform encephalopathy (BSE) in cattle, and the subsequent human cases of a then novel variant of Cruetzfeldt-Jakob disease (vCJD) linked to the bovine disease, led to some of the defining political moments of the time and large-scale reassessment of agricultural practices and food safety. From the then Agriculture Minister, John Gummer, giving his daughter a hamburger to reassure the nation that British beef was safe in 1990, to the mass slaughter of over 4 million cattle to contain the BSE epidemic, images from the period still resonate in the country's consciousness. Given the lingering shadow of the BSE and vCJD outbreaks, the news, as reported by David Holmes in this month's Newsdesk, that so far in 2012 not a single case of the human disease has been reported in the UK is particularly welcome. 2012 looks like being the first year since 1995 without a reported case.

Kidney transplants performed in Europe are considerably more successful in the long run than those performed in the United States. While the one-year survival rate is 90% in both Europe and the United States, after five years, 77% of the donor kidneys in Europe still function, while in the United States, this rate among white Americans is only 71%. After ten years, graft survival for the two groups is 56% versus 46%, respectively. The lower survival rates compared to Europe also apply to Hispanic Americans, in whom 48% of the transplanted kidneys still function after ten years, and particularly to African Americans, whose graft survival is a mere 33%.
